I think it was a double meaning - the revelation that one of the Losties was going to be Jacob's substitute on-island, and also that alt-reality Locke was becoming a substitute teacher.

I wouldn't be surprised if if has something to do with the names in the cave. Maybe it refers to the search for a substitute guardian of the island.
